How you use the feature is up to you. You can easily modify a custom group via the Custom Groups tab in Membership Management.
You certainly should have members permission to assign them as mentors. You can use getting CL credit as a motivator.
Note that the built-in mentor groups only work for those mentors that are assigned. If you have some that are not assigned they will not show up in the built in groups.

I guess, I am not sure of how the feature works - that 's why are these questions.. 
What would be an email address of the newly created NSTM Mentors group, if any?
For the default lists, is it only including people already assigned as mentors/and those with assigned mentors?

An email address is automatically created for the group. It will be in the form
groupname-nnnn@toastmastersclubs.org,
where "groupname" is the name of the group and nnnn stands for your club number.
For your groupname you could use something like mentorpool The email address would then be mentorpool-nnnn@toastmastersclubs.org
